چکیده

Abstract Background 3-dimensional (3D) cultured tonsil-derived mesenchymal stem cells (TMSCs) showed higher therapeutic efficacy in murine colitis model than conventional 2D-cultured TMSCs via increased anti-inflammatory cytokine expression. Here, we evaluated the in-vivo localization and formation of 3D-cultured using electron microscopy. Methods Chronic mouse was induced by oral administration 1.5% dextran sulfate sodium for 30 days. injected intraperitoneally on day 6 16 after inducing mice. TMSC as a spheroid 24 hours before intraperitoneal injection. On 31, mice were euthanized immunofluorescence analysis. Moreover, characterization Results In vitro spheroids measured to have an average size 182.4 μm 1, 169.4 2, 154.1 3; however, gradually decreased 2 days culture. treated with TMSCs, there no detectable at time sacrifice. white variable sized spherical aggregates observed. These attached peritoneal cavity including omentum mesentery. fluorescence microscopy, these stained human nuclear antigen antibody, indicating that transplanted formed cluster peritoneum even 14 covered lymphocytes. inside spheroids, suspected cell mesothelial distributed. Conclusion Intraperitoneally found Based results current study, functional studies evaluating role therapy, cell-to-cell communication between immune cells, are needed.

Immune Suppressive Effects of Tonsil-Derived Mesenchymal Stem Cells on Mouse Bone-Marrow-Derived Dendritic Cells

Mesenchymal stem cells (MSCs) are considered valuable sources for cell therapy because of their immune regulatory function. Here, we investigated the effects of tonsil-derived MSCs (T-MSCs) on the differentiation, maturation, and function of dendritic cells (DCs). We examined the effect of T-MSCs on differentiation and maturation of bone-marrow- (BM-) derived monocytes into DCs and we found sup...

Study of Chondrogenic Effects of Chondrocytes Cocultured With Murine Bone Marrow-Derived Mesenchymal Stem Cells

Purpose: Co-culture systems of marrow derived mesenchymal stem cells (mMSCs) with mature chondrocytes have theoretically been considered as a putative way of MSCs chondrogenic differentiation. MSCs differentiated in this system could be used for transplantation purpose without of any need to their purification since the cells with which MSCs are co cultured are native cartilage cells. Despite o...

Therapeutic potential of tonsil-derived mesenchymal stem cells in dextran sulfate sodium-induced experimental murine colitis

The therapeutic potential of tonsil-derived mesenchymal stem cells (TMSC) prepared from human tonsillar tissue has been studied in animal models for several diseases such as hepatic injury, hypoparathyroidism, diabetes and muscle dystrophy. In this study, we examined the therapeutic effects of TMSC in a dextran sulfate sodium (DSS)-induced colitis model. TMSC were injected in DSS-induced coliti...

Sarcoma derived from cultured mesenchymal stem cells.

To study the biodistribution of MSCs, we labeled adult murine C57BL/6 MSCs with firefly luciferase and DsRed2 fluorescent protein using nonviral Sleeping Beauty transposons and coinfused labeled MSCs with bone marrow into irradiated allogeneic recipients. Using in vivo whole-body imaging, luciferase signals were shown to be increased between weeks 3 and 12. Unexpectedly, some mice with the high...
